Tissue engineering represents a promising strategy for the regeneration of damaged
cartilage. This strategy is based on cell-scaffold systems in which the
scaffold serves as a temporary extracellular matrix (ECM) to promote cartilage
regeneration. In-situ forming hydrogels are attractive scaffold candidates
because they provide the essential 3D environment for chondrocytes to
proliferate and differentiate meanwhile allowing transportation of nutrients
and waste products. In addition, they allow minimally invasive techniques and
easy encapsulation of chondrocytes.
In recent studies, injectable hydrogels were prepared by the horseradish peroxidase-mediated
co-crosslinking of dextran-tyramine (Dex-TA), hyaluronic acid-tyramine and
heparin-tyramine (Hep-TA) conjugates and used as scaffolds for cartilage tissue
engineering. Based on these results several studies were performed to evaluate
the potential of the material to be used for in-vivo application. Therefore a
first study was performed on horse cadavers. The results will be presented
during the meeting.
The results revealed that enzymatically crosslinked hydrogels can induce chondrocyte viability and
proliferation, and an enhanced matrix production. Adhesion tests reveal the
adherence of the gel to cartilage if a primer is used. The chirurgical
procedures have been optimized for future experiments. The results indicate
that these hydrogels have a high potential as matrices for cartilage tissue regeneration.
